.mkv files are makarovska files and are for watching movies in HD on the PC.
For them to work on the xbox 360, you will have to convert them.

As the guys were saying, before india tried to rubbish them off incorrectly, the xbox doesn't support all formats, or all codecs like the ps3 does.
I'm pretty sure divx is supported, so use that converter, when it's done, put the file on USB and plug in.
However i'm not sure if it will come out in HD. Unless you can convert it to a resolution that is in HD with the program.

are you using windows 7? if so you can just use windows media center to stream anything onto your xbox including mkv if you install this on your pc http://www.divx.com/en/software/divx-plus/codec-pack . all you should have to do in windows media center on your pc is to add your external hardrive as one of your sources of media and you should be good to go.
setting up the connection between your pc and the xbox 360 via media center should be straight forward as it should tell you what to do. (assuming everything is on the same network).
